Resorption of Dentin by Osteoclast-Like Cells Differentiated From Calcium-Deficient Mouse Bone Marrow Cells in the Presence of Tumor Necrosis Factor-¥á in Vitro

|
The purpose of this study was to investigate the differentiation of osteoclast-like cells from calcium-deficient mouse bone marrow cells and the resorption of dentin by those in the presence of rTNF-a. Bone marrow cells were prepared from tibia
of
mice
fed with the low calcium diet for 2 weeks, and the collected bone marrow cells were cultured on dentin slices and holed dentin specimens for 6 days in the presence of rTNF-a(final concentration : 100ng/ml). In the holed specimens, rTNF-a was
applied to
the hole, which could diffuse to the medium through dentinal tubules. After cultures, we examined the morphology of the osteoclast-like giant cells and resorption pits on dentin slices and holed dentin specimens by means of scanning electron
microscopy,
and observed tartrate-resistant acid phosphatase(TRACP) activity of the differentiated cells on the dentin slices. The results showed that osteoclast-like giant cells and resorption pits were identified on the surface of dentin slices and holed
specimen
in the SEM views, TRACP-positive mononuclear and multinucleated giant cells were also found. The size and shape of resorption lacunaes were variable and the contour of osteoclast-like cells was irregular. Osteoclast-like cells showing many
filopodia on
the functional surface and short microvilli on dorsal surface were found. Fusion between the round mononuclear cells and giant cells was observed.
